Donors are mobilized with G-CSF, plerixafor, or G-CSF and plerixafor prior to collection. Isolated from leukapheresis samples using density gradient separation or red blood cell lysis and cryopreserved in CryoStorCS10 medium, cells are obtained using Institutional Review Board (IRB)-approved consent forms and protocols. Additional documentation and high-resolution HLA typing (Class I and Class II alleles and CMV status) are available upon request. ACDA is added as an anticoagulant. This cell line uses EFC technology to measure ligand-induced internalization of PRLHR via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged PRLHR. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting PRLHR-arrestin complex is internalized to the early endosome, leading to production of EFC signal.
